Proposed Interstate 326 would follow the freeway section of U.S. 321 from Interstate 26 in Johnson City east to Elizabethton. While the route is already a freeway, the interchange between Interstate 26 and U.S. 321 (Milligan Highway) would have to be upgraded to Interstate standards.

Interstate 326 is a designation proposed by the Johnson City Metropolitan Transportation Planning Organization for the U.S. 321 freeway from Interstate 26 and U.S. 23 in Johnson City east to Elizabethton. On the MTPO web page, the description of Interstate 26 Extension includes a proposed spur route: "I-326 possible number for US 321 (SR 67) Existing freeway between Johnson City and Elizabethton, TN." There is no additional information on the timing of upgrading this section of U.S. 321 to Interstate standards.
